UV-Vis Spectrophotometer (Saleh lab)
- Start up the computer and the UV-Vis using the button in the lower left corner, and open the UV-Vis application on the computer. This will take a couple minutes, so you can prepare your cuvette while it's going.
- Very carefully and with gloved hands, clean the exterior of the cuvette with a Kim wipe and rinse the inside with DI water.
- Fill the cuvette with DI water. This is for a blank run.
- Place the cuvette into the holder with the unfrosted side facing the incident light and lock it in with the lever on the left.
- Once the UV-Vis app has started up, click on the icon for both light bulbs and select "Light ON". If the machine is not fully warmed up, the "Light ON" option will be grayed out but should be clickable in a few minutes.
- Once both lights are ON, click the "Blank" button.
- After the blank is done running, a reading will pop up on the screen. If the reading is too noisy, clean the cuvette, and run it again. Once a good blank has been achieved, prepare your sample.
- If you have enough sample to fill the cuvette multiple times, fill the cuvette with your sample and dump it out. This is to prevent any dilution that may happen if there is still DI water in the cuvette.
- Then fill the cuvette again with your sample and lock it into the holder.
- Click the "Setup" button and enter your target wavelength.
- Click the "Sample" button and wait for the Magic™.
- Your reading will pop up, and the intersection with the target wavelength will appear below the reading.
- Clean the cuvette with DI water and place on a Kim wipe to dry.
- Turn off the UV-Vis.
